Introduction
Proper storage and handling of peptides is essential to maintaining their structural integrity and biological activity throughout a research study. Even high-purity peptides can degrade rapidly if exposed to incorrect temperatures, light, moisture, or repeated freeze-thaw cycles.
This guide covers best practices for storing both lyophilised (freeze-dried) and reconstituted peptide solutions. All products supplied by Alpha Peptides are intended strictly for in vitro and laboratory research use only.
Lyophilised Peptide Storage
Lyophilised peptides are the most stable form and should be your default storage format until the point of use. General guidelines:
- Short-term storage (up to 3 months): Store at −20°C in a standard laboratory freezer. Ensure the vial is sealed and desiccated.
- Long-term storage (3 months to 2+ years): Store at −80°C for maximum stability. This is particularly important for larger, more complex peptides prone to oxidation.
- Avoid repeated temperature cycling: Each time a frozen vial warms to room temperature, moisture can enter and begin degrading the peptide. Only remove vials from storage when ready to reconstitute.
- Keep away from light: UV exposure can cause photodegradation, particularly in peptides containing tyrosine, tryptophan, or phenylalanine residues. Store in amber vials or opaque containers where possible.
- Desiccation: Store vials with a desiccant packet to absorb any ambient moisture, especially in humid laboratory environments.
Reconstituted Peptide Solution Storage
Once a peptide has been reconstituted, its stability window is significantly shorter. Follow these guidelines to maximise usable life:
- Refrigerate at 2–8°C immediately after reconstitution. Do not leave reconstituted solutions at room temperature for extended periods.
- Do not freeze reconstituted solutions. Ice crystal formation during freezing can physically damage peptide structure. If long-term storage of a reconstituted solution is required, aliquot into single-use volumes before freezing to avoid repeated freeze-thaw cycles.
- Use within 28–30 days when reconstituted with bacteriostatic water (0.9% benzyl alcohol), which acts as a preservative.
- Use within 24 hours when reconstituted with sterile water for injection (WFI), which contains no preservative.
- Protect from light: Wrap vials in foil or use amber glass vials for light-sensitive peptides.
Understanding Freeze-Thaw Cycles
Repeated freeze-thaw cycles are one of the most common causes of peptide degradation in research settings. Each cycle subjects the peptide to mechanical stress from ice crystal formation and thawing, as well as concentration effects as water freezes out of solution.
Best practice: If you need to store a reconstituted peptide for longer than the recommended refrigerated window, aliquot the solution into single-use volumes (e.g., 0.1–0.5 mL per vial) before freezing. This way, each aliquot is only thawed once before use.
Peptide Stability Factors
Several intrinsic and extrinsic factors affect how quickly a peptide degrades:
- Sequence composition: Peptides containing methionine, cysteine, or tryptophan are more susceptible to oxidation. Those with asparagine or glutamine may undergo deamidation over time.
- pH: Most peptides are most stable at a slightly acidic pH (4–6). Alkaline conditions can accelerate hydrolysis.
- Temperature: Every 10°C increase in temperature roughly doubles the rate of chemical degradation (Arrhenius principle). Cold storage is always preferable.
- Oxygen exposure: Minimise headspace in vials and consider using nitrogen or argon gas to purge vials before sealing if working with oxidation-sensitive peptides.
Handling Best Practices
- Always wear nitrile gloves when handling peptide vials to prevent contamination from skin oils and proteases.
- Use sterile, single-use syringes and needles for each withdrawal from a vial.
- Allow frozen or refrigerated vials to reach room temperature before opening to prevent condensation entering the vial.
- Label all vials clearly with: peptide name, concentration (if reconstituted), date of preparation, and expiry date.
- Maintain a storage log for each peptide, recording batch number, COA reference, storage conditions, and any observed changes in appearance.
Signs of Peptide Degradation
Discard any peptide sample that shows the following:
- Cloudiness or visible particulates in a reconstituted solution that was previously clear
- Unexpected colour change
- Unusual odour upon opening a lyophilised vial
- Clumping or discolouration of the lyophilised powder
