Introduction
Peptide reconstitution is one of the most critical steps in any research protocol. Lyophilised (freeze-dried) peptides are stable for long-term storage, but must be carefully reconstituted before use. Errors at this stage — using the wrong solvent, introducing contamination, or miscalculating concentration — can compromise the integrity of your entire study.

What You Will Need
- Lyophilised peptide vial
- Bacteriostatic water (BW) or sterile water for injection (WFI)
- Alcohol swabs (70% isopropyl)
- Sterile syringes (1 mL insulin syringes are commonly used for precision)
- Sterile needles (23–25 gauge)
- A clean, low-particulate workspace
Solvent Selection: Bacteriostatic Water vs Sterile Water
- Bacteriostatic water (0.9% benzyl alcohol) — preferred for most peptides. Extends usable life to approximately 28–30 days when refrigerated at 2–8°C.
- Sterile water for injection (WFI) — suitable when the entire reconstituted volume will be used immediately. Use within 24 hours.
- Acetic acid solution (0.1–1%) — required for certain peptides poorly soluble in water alone. Check the product COA for guidance.
Step-by-Step Reconstitution Protocol
Step 1 — Prepare Your Workspace
Work in a clean environment, ideally near a laminar flow hood. Wash hands thoroughly and wear nitrile gloves throughout.
Step 2 — Allow the Vial to Reach Room Temperature
If stored frozen, allow the vial to equilibrate to room temperature before opening to prevent condensation and moisture absorption.
Step 3 — Disinfect the Vial Septum
Wipe the rubber septum of both the peptide vial and solvent vial with a fresh alcohol swab. Allow to air-dry for 30 seconds.
Step 4 — Draw Up the Solvent
Using a sterile syringe and needle, draw up the required volume of bacteriostatic water. The volume determines your final concentration — see the calculation section below.
Step 5 — Inject the Solvent Slowly
Insert the needle through the septum at an angle, directing the stream of solvent against the glass wall rather than directly onto the lyophilised powder. This prevents foaming and degradation.
Step 6 — Allow to Dissolve — Do Not Shake
Gently swirl or roll the vial between your palms until fully dissolved. Never vortex or shake vigorously — this can cause aggregation and denaturation. Some peptides may take several minutes to dissolve fully.
Step 7 — Inspect the Solution
The reconstituted solution should be clear and free of particulates. If cloudy or containing visible particles after thorough mixing, do not use — the peptide may have degraded or the solvent may be incompatible.
Concentration Calculation
Concentration (mg/mL) = Peptide mass (mg) ÷ Volume of solvent added (mL)
Example: A 10 mg vial reconstituted with 2 mL of bacteriostatic water yields 5 mg/mL (5,000 mcg/mL). Each 0.1 mL drawn contains 500 mcg.
Storage After Reconstitution
- Refrigerate at 2–8°C. Do not freeze reconstituted solutions.
- Keep away from light — use amber vials or wrap in foil for light-sensitive peptides.
- Use within 28–30 days with bacteriostatic water, or 24 hours with sterile water.
- Label vials with peptide name, concentration, reconstitution date, and expiry.
Common Mistakes to Avoid
- Injecting solvent directly onto the powder — always aim at the glass wall.
- Shaking the vial — swirl gently only.
- Using tap water or saline — not appropriate solvents for peptide reconstitution.
- Reconstituting too far in advance — only reconstitute what you need for the study period.
- Skipping room-temperature equilibration — can introduce moisture and affect solubility.
